3 ДНК метилирование
ферментативный перенос метилтрансферазами (см. ДНК-метилазы) метильной группы (-CH3) от S-аденозилметионином (см. S-аденозилметионин) на молекулу ДНК. ДНК прокариот содержит модифицированные основания N 6-метиладенин и 5-метилцитозин, тогда как ДНК высших эукариот — в основном 5-метилцитозин. При этом метилируется цитозин, расположенный в динуклеотиде ЦфГ. У эукариот М. ДНК обычно ассоциировано с инактивацией экспрессии генов (см. молчание генов). Благодаря существующей в бактериях системе метилирования-рекогниции клетки способны идентифицировать свой генетический материал и отличать его от инородных молекул, проникших в клетку тем или иным способом: процесс М. сайтов рестрикции (модификация) предохраняет бактериальную ДНК от разрушения собственными эндонуклеазами. У млекопитающих ДНК м. приводит к стабильному подавлению экспрессии части генов (см. молчание генов). ДНК м. играет также важную роль в инактивации одной из Х-хромосомы у самок, в регуляции экспрессии генов в процессе развития, а также непосредственно вовлечено в феномен геномного импринтинга (см. импринтинг (2)), связанного с различной пенетрантностью некоторых аллелей в зависимости от их происхождения, т.е. прохождения через материнский или отцовский гаметогенез.
